(how) is it possible to store a file from my website’s backend on my nextcloud?
E.g. by using an accesslink?

It should be possible using WebDAV.
Google search: webdav linux nextcloud - Google Suche
Connect from your website to Nextcloud using WebDAV and copy/move the files to Nextcloud, the files will be listed under Nextcloud then.
Perhaps you could clarify how you are hosting your website, along with whether it is on the same machine as your Nextcloud. Thanks.
